A subunidade DGCR8 do complexo microprocesador (iniciais en inglés de DiGeorge syndrome chromosomal [ou critical] region 8, ou "síndrome de DiGeorge cromosómica [ou crítica]] rexión 8") é unha proteína que nos humanos está codificada no xene DGCR8 do cromosoma 22, que colabora no procesamento de miARN durante a interferencia de ARN.[1] Aínda que en mamíferos onome que se lle dá é DGCR8, noutros animais como os organismos modelo Drosophila melanogaster e Caenorhabditis elegans, esta proteína coñecese co nome de Pasha (colaboradora de Drosha).[2] En todos eses animais é un compoñente necesario da vía de interferencia de ARN.
Función
DGCR8 is localized to the cell nucleus and is required for microRNA (miRNA) processing. It binds to Drosha, an RNase III enzyme, to form the Microprocessor complex that cleaves a primary transcript known as pri-miRNA to a characteristic stem-loop structure known as a pre-miRNA, which is then further processed to miRNA fragments by the enzyme Dicer. DGCR8 contains an RNA-binding domain and is thought to bind pri-miRNA to stabilize it for processing by Drosha.[3]
